Background
c-Kit is a transmembrane tyrosine kinase encoded by the cKit proto-oncogene. It regulates various biological responses such as cell proliferation, apoptosis, chemotaxis, and adhesion. Ligand binding to its extracellular domain triggers autophosphorylation on several tyrosine residues within its cytoplasmic domain, leading to its activation. Mutations in c-Kit have been implicated in tumor growth and progression in multiple cancers, including mast cell diseases, gastrointestinal stromal tumors, acute myeloid leukemia, Ewing sarcoma, and lung cancer. Phosphorylation at tyrosine 721 of c-Kit facilitates the binding and activation of PI3 kinase.
